Cibler les ions cuivre pour les éliminer ou les redistribuer est actuellement considéré comme une stratégie thérapeutique possible dans le contexte de la maladie d’Alzheimer (MA). Les paramètres thermodynamiques, comme par exemple l’affinité pour le cuivre (II) ou la sélectivité du cuivre (II) par rapport au zinc (II), sont pris en compte dans la conception d’un éventuel traitement. En revanche, les paramètres cinétiques ont été négligés malgré leur importance probablement élevée. Dans la présente communication, nous utilisons une série de ligands azamacrocycliques pour démontrer que les problèmes cinétiques doivent être pris en compte lors de la conception de candidats-thérapeutiques ciblant le cuivre dans le contexte de la MA.

Targeting copper ions to either remove or redistribute them is currently viewed as a possible therapeutic strategy in the context of Alzheimer’s disease (AD). Thermodynamic parameters, as for instance the copper(II) affinity of the drug candidate or the copper(II) over zinc(II) selectivity, are considered in the design of the drug candidate. In contrast, kinetic ones have been overlooked despite their probable high importance. In the present communication, we use a series of azamacrocyclic ligands to demonstrate that kinetic issues must be taken into account when designing copper-targeting drug candidates in the context of AD.
